Shut-down
17. At end of your session (or anytime you wish to change slides during your session), repeat steps 14b, c,
and g, except now to remove a slide from the stage. Leave either the 5X or 10X objective lens in place
for the next user. If you used any fluid-immersion lens, clean the lens by wicking with lens paper as
demonstrated during training (remember: your fingers should not apply pressure to the lens at all).
18. Because the confocal computer may crash at any time, take a copy of your files with you. These can be
burned on a CD or copied to a portable USB drive (e.g. flash drive).
19. Check sign-up sheet on confocal room door. Will another user be using the scope within 2 hr? If yes, go
to step 20. If no, go to step 21.
20. (Next user is coming on within 2 hr.)
a. Any lasers (and Hg lamp) that are on should be left on unless next user has told you s/he will not be
using a particular laser. If the Argon laser is on, with Argon/2 selected, click Standby button.
b. With File selected, click Exit button.
c. Click Exit button on Switchboard window (see screenshot
accompanying step 8). A message pops up if one or more lasers are
on or in standby. Click OK.
d. In the lower right corner of monitor, a message will appear,
followed by another.
This typically occurs
3 times, but the first
2 come in quick
succession and are not always clearly displayed. Wait for
the 3 occurrences of this pair of messages to be displayed
(if you are not sure if all 3 have occurred, just wait about
30 seconds after seeing the first message).
e. Click Start button in lower left corner of monitor,
select Shut Down, and then, when Shut Down Windows
dialog box appears, select the Log off option.
f. Fill out the log book. You can now leave.
