c. Click Channels.

d. The number of Channels buttons shown is the number of fluors

that will be imaged (3 in this example). Click the button of the

fluor with the longest maximum emission wavelength

e. Click 1 button on Pinhole line.

Note the Optical slice thickness.

f. Click the Channels button of the fluor with the next longest

maximum emission wavelength.

g. Adjust Pinhole slider so that Optical slice thickness for second

fluor is the same as for the first fluor (from step 13e).

h. Repeat steps 13f-g if a third fluor is being imaged.

14. Loading sample slide on microscope stage.

a. Tap Microscope button on microscope’s touch screen.

b. Have 5X or 10X objective lens in place before loading a slide on

the stage. If desired lens is not in place, select it by tapping its

button (when Objectives tab is selected).

 Always switch objectives using software (on monitor or touch screen), NEVER switch

objectives by hand.

c. To prevent accidentally hitting an objective lens while loading slide onto stage, take one or both

precautions:

Using joystick, move stage forward so that depression that receives

slide is as far forward from objective lenses

as possible.

 Joystick has coarse and fine movements. Toggle

between them by pushing straight down on joystick until

you feel a small click.

Tap Load position

(stage will drop).
